DNA Manipulation

 

DNA cloning and sequence manipulation are essential tools in all life science disciplines.  The Restriction Free (RF) and Transfer-PCR (TPCR) cloning methodologies are gaining momentum as alternative approaches to the traditional cloning methods, which are based on restriction endonucleases followed by a ligation step. Since their implementation, the applications of the RF/TPCR methodologies have been extended beyond their original utilization for single gene cloning. The strength of these methods lies in their simplicity and flexibility, which can be harnessed for a large spectrum of applications. These include cloning of multiple DNA fragments, insertion and/or deletion of specific DNA sequences, and single and multiple-site mutagenesis.
