Director of Research at CNRS, Co-founder and Scientific Advisor at PicoSeqOrganizer Department of Biomolecular SciencesContact Abstract Show full text abstract about We have developed a novel method for DNA detection and seque...» We have developed a novel method for DNA detection and sequencing based on the mechanical opening and closing of single DNA hairpins and the detection of roadblocks to rehybridization due to the presence in solution of complementary fragments. Using small magnets to pull on single DNA hairpins tethering magnetic micro-beads, the position of these roadblocks can be detected with nanometer precision. That signal provides a unique finger-print which can be used to identify and quantify DNA molecules in a sample. The approach is sufficiently general and robust to allow for DNA sequencing by hybridization or by ligation. We will present new results pertaining to telomere sequencing and the detection of methylation patterns on single DNA molecules.
